Biological Efficiency Testing According ISO 14698-1 | Author : Roberto Ligugnana*,Roberto Montagna | Abstract | Full Text | Abstract :The biological collection efficiency of the active multi-nozzle impactor air sampler TRIO. BAS MINI was compared to the efficiency of the slit to agar air sampler called CASELLA type that is used as traditional reference air sampler device. The TRIO.BAS biological efficiency was 82% vs Casella microbial air sampler [1,2]. Considering that the evaluation of biological efficiency between different microbial air samplers is always difficult and relative, the two results can be considered statistically comparable. |
| Highlights of CRISPR-Cas9 Genome Editing | Author : Ali Movahedi*, Amir Almasizadehyaghuti, Kourosh Mohammadi, Sirus Sangari, Qiang Zhuge | Abstract | Full Text | Abstract :The advantage of CRISPR genome editing is not limited to only its cost, but it is easy to use in any lab with molecular biology expertise. On the other hand, CRISPR does not need to protein engineering for targeting each gene, while TALE and ZF nucleases are needed. CRISPR-Cas9 genome editing system needs only a DNA construct to encode the target specific gRNA and Cas9 in one expression vector. Furthermore, CRISPR-Cas9 can edit multiple genome with homolog sequencing simultaneously, resulting in increasing the efficiency. |
| Presence of zoonotichelminths in cats (Feliscatus) of condominiums in City of Mexico with special concern to Toxocara cati infection | Author : Ignacio Martínez-Barbabosa*,Rodrigo de Jesús Pimienta Lastra, Hilda Ortiz Pérez, José Rodolfo Marcos Aguilar Venegas, Ana María Fernández Presas | Abstract | Full Text | Abstract :The toxocariasis is a soil-transmitted helminthiasis (SHT) of worldwide distribution, caused by the intestinal helminths of the dog and the cat Toxocaracanis and Toxocaracati, respectively. The toxocariasis by T. cati is little studied worldwide.
Objective: Determine the prevalence of T. cati infection and other intestinal zoonotic parasites in domestic cats that live in condominium apartments in Mexico City.
Material and Methods: With Faust’s method, 121 feces of cats living in apartments in a condominium in Mexico City were analyzed. A questionnaire was applied to the owners to obtain hygienic habits of the animal. The statistical analysis was performed with the Chi-square and Fisher’s exact tests with a confidence level of p <0.05.
Results: The prevalence of T. cati infection was 33.1%. Embryonated eggs were observed in 8.26% p < 0.000; oocysts of Toxoplasma gondiiin 6.6% and eggs and larvae of ancylostomatidsin 1.6%. The association of Ancylostoma spp. and T. gondii sp. was significant (p < 0.004). The highest frequency of infection was found in cats under 12 months of age. However, the presence of T. cati in the different age groups was p < 0.000.
Conclusion: The coexistence with parasitized animals is a risk to acquire different intestinal zoonoses transmitted by the cat as toxocariasis. However, the presence within the home of larvae eggs of T. cati and oocysts of T. gondii increases the risk of infection in younger children and people suffering from some type of immune deficiency. |
| Influence of Coating Developed from Oligomer Isolated from Lac Resin on Post-Harvest Quality and Shelf Life of Peaches (Prunus Persica L.) | Author : Anjali Bishnoi, Chawla HM, Maulin P Shah* | Abstract | Full Text | Abstract :Large quantities of fresh fruits are produced that never reach the consumers due to heavy post-harvest losses, lack of storage, transportation care & less acceptable quality. These losses are not only concerned in terms of revenue but it also concern in terms of health, life style and environment. To full fill consumer demand and environmental, several methods are being utilized to reduce these losses of fruits. Fruit coatings are considered as one of the widely used methods. This work investigates the effect of fruit coating developed from oligomer (P-104), isolated from lac resin on quality of peach (Prunus persica L.) when stored at room temperature (38 ± 2 °C) and at 4 °C temperature. Fruit quality was evaluated by measuring physiological weight loss, color and textural changes as well as microbiological evaluation at a regular interval of four days. When kept at room temperature, uncoated peaches remained fresh and microbiologically safe for 4 days only while shelf life of coated peaches increased to 12 days. Again the shelf life of coated peaches prolonged to 24 days when stored at 4°C. These results showed that combine effect of coating and low storage temperature could improve shelf life of peaches, indicating that coating and low temperature preservation could be a potential method for fruits preservation. Thus, this study suggests that fruits may be coated and preserved at 4°C to ensure the health, life style, economic and environment by extending the shelf life of peaches. |
| Open Reading Frame Filtering Methods for Identification of Genic Sequences | Author : Nayra M. Al-Thani, Simeon S. Andrews, Dietrich Büsselberg* | Abstract | Full Text | Abstract :Protein-Protein Interactions (PPIs) help understanding disease processes and their mechanisms. Ideally, researcher would like to understand the full network of PPIs that take place within a cell “interactome”, that is why Open Reading Frame (ORF) filtering method was utilized. ORF is a DNA fragment that lacks stop codon and has the potential to demonstrate a physiological interaction. ORFs are obtained by random shearing of DNA into small fragments. Fragments are filtered by insertion upstream of a selectable marker, to allow the survival of cells that only have an ORF. As vast majority of out of frame fragments encodes a premature stop codon. Here we present a method for filtering of genic ORFs ‘real gene’ which results in a physiological protein. Furthermore, researchers thought fragments libraries rather than full-length libraries are perhaps counterintuitive, as expected to result in a high rate of false negatives. However, researchers have found fragments libraries rather than full-length reduce number of false negative interactions. Lastly, great advantage to note is while using fragmented library it allows localization of interaction site, offering a robust path for drug targets, treatments towards cancer and the arising resistance to antibiotics. |
