| Creation of initial breeding material of potato with complex resistance to Fusarium dry rot and tuber late blight | Author : Valentyna Hordii?nko; Nataliia Zakharchuk | Abstract | Full Text | Abstract :Purpose. To select the initial breeding material with complex resistance to Fusarium dry rot and tuber late blight among the created potato of secondary interspecific hybrids.
Methods. Interspecific hybridization, laboratory test, analytical approach.
Results. Based on the interspecific hybridization, the initial breeding material was created and the degree of its resistance to the above pathogens was determined by way of artificial infection of tubers with the inoculum of such fungi as Fusarium sambucinum Fuck and Phytophthora infestans (Mont.) De Bary. During interspecific hybridization based on schemes of saturating and enriching crosses, using forms of various species with a high phenotypic expression of resistance to Fusarium dry rot, the result of the cumulative effect of genes that control resistance to the pathogen was observed. Crossing combinations differed significantly for the degree of population average manifestation of resistance to the diseases.
Conclusions. Combinations ?54, ?53, ?61 with a mean resistance (above 7 grades) to Fusarium dry rot have been selected. Such combinations as B52, B50 and B54 had increased resistance to tuber late blight. It was found that the combination ?54 is characterized by complex resistance to both diseases. For further work, the following samples with complex resistance to Fusarium dry rot and tuber late blight (7 grades or more) were selected: ?59?42, ?59?43, ?50?16, ?50?19, ?50?44, ?51?1, ?51?26, ?51?28, ?52?11, ?52?23, ?52?24, ?52?29, ?53?1, ?53?11, ?53?17 , ?53?23, ?54?13, ?54?14. |
| Breeding and genetic peculiarities of modern spring barley varieties for grain number per main ear | Author : Stanislav Vasylkivskyi; Volodymyr Hudzenko; Oleksandr Demydov; Olha Barban; Svetlana Kolyadenko; Ivanna Smulskaya | Abstract | Full Text | Abstract :Purpose. To reveal breeding and genetic peculiarities of modern spring barley varieties for the “number of grains per main ear” trait and identify genetic sources of increased combining ability for involving in hybridization.
Methods. Investigations were carried out at the V. M. Remeslo Myronivka Institute of Wheat of NAAS of Ukraine. Modern varieties of domestic (‘Virazh’, ‘Talisman Myronivskyi’, ‘Komandor’) and foreign (‘KWS Aliciana’, ‘KWS Bambina’, ‘Zhana’, ‘Explorer’) breeding were involved in crossing for a full (7´7) diallel scheme. Parents and F1 were studied in field conditions during 2014–2016.
Results. The analysis of variance of combining ability has shown a significant advantage in varying of general combining ability (GCA) effects. The mean square of specific combining ability (SCA) was significantly less than the GCA, but reliable throughout the years. The reciprocal effect was reliable only in 2014. Stably high effects of GCA during all years of investigations were noted in the varieties ‘KWS Aliciana’ (1.18–1.62) and ‘Virazh’ (1.33–1.48). The variety ‘KWS Bambina’ was characterized by lower but reliable positive effects of GCA (0.43–0.99) as compared to mentioned above. Non-allelic gene interaction was not found, that allowed to calculate the basic parameters of genetic variation. During all years of investigations, dominant effects of genes (H1 and H2) prevailed over the additive (D) ones in phenotypic expression of grain number per main ear. Mean degree of dominance in the experiment (H1/D) has shown overdominance. The same pattern was also distinctive for the index of mean degree of dominance in the loci . The dominance was reliably directed. Dominant effects of genes increased grain content, and recessive ones reduced it. At least 3–4 genes (groups of genes) have been revealed which determined the effects of dominance. At the same time, recessive genes (F<0) or gene effects were prevailed quantitatively in the varieties investigated. A high coefficient of heritability in broad sense (H2 = 0.98) has shown a significant determination of phenotypic variability with genetic factors. The coefficient of heritability in narrow sense (h2 = 0.66–0.68) confirmed that despite the advantage of dominant effects over the additive ones, the contribution of the latter was also significant.
Conclusions. The prevalence of dominant effects of genes in the phenotypic expression of the number of grains per main ear causes the need for sufficient sample size of hybrid material and points to the expediency of conducting a more “rigid” selection for phenotype in later generations. At the same time, the considerable contribution of additive effects and high values of heritability indices give reason to predict the efficiency of selections aimed at increasing the trait in created hybrid material. The varieties ‘Virazh’, ‘KWS Aliciana’, ‘KWS Bambina’ should be used as effective genetic sources to increase grain content in combination breeding |
| Dynamics of correlation relationship changes in new winter wheat varieties (Triticum aestivum L.) under the influence of environmental factors in the Southern Forest-Steppe zone of Ukraine | Author : Havryliuk, M. M.; Kalenych, P. Ye. | Abstract | Full Text | Abstract :Purpose. To establish correlation dependence of influence of such factors as seeding rate, sowing time and seeding methods on yield indicators, as well as individual response of winter wheat varieties to them in case of the ecological system of seed production, that would allow to form elements of intensive varietal technology and obtain the genetically determined productivity potential of the variety in the future.
Methods. Weight measuring was used to determine seed yield; the reliability of the experiment results was identified by statistical evaluation, analysis of variance and regression procedure.
Results. During the period of investigation, the weather conditions differed significantly both for the temperature regime and the amount of precipitation, but the stability of grain and seed yield over years was the main requirement to varieties, that allowed to study the intergenotypic correlation relationships of crop yield indices and the dynamics of their changes under the effect of ecological factors. In the course of investigation, winter wheat yield indices were fixed annually from 684 plots, which were grouped and analyzed for factors of influence in order to ensure the complete certainty.
Conclusions. Variants of investigation were defined for the certified seed yield and environmental factors (sowing time and seeding method, seeding rate), that had strong positive correlation relationships. Seeding rate and seeding method had the greatest impact on 1000 kernel weight. For tillering coefficient, varieties depending on the variant of the investigation had a negative close correlation relationship, which was greatly affected by seeding rate. For correlation relationships, winter wheat varieties ‘Bohdana’ and ‘Slavna’ appeared to be the most stable, ‘Chorniava’ and ‘Astarta’ were the most plastic. |
| Method of propagation, stimulation of rhizomes growth in vitro culture and adaptation in the open ground for the genus Miscanthus representatives | Author : Svitlana Gontarenko; Snizhana Lashuk | Abstract | Full Text | Abstract :Purpose. To develop a method of propagation, stimulation of rhizomes growth in vitro culture for the genus Miscanthus representatives and their adaptation in the open field without the use of greenhouse complexes for acclimatization and completion of growing.
Methods. Biotechnological procedures, mathematical and statistical analyses.
Results. Prescription of nutrient medium was developed for explants inoculation, sprouts propagation, rhizomes growth stimulation in vitro. Such sterile explants as seeds, buds to be removed from rhizomes, parts of stems with bud were placed on modified media with mineral portion by Murashige and Skoog (MS) that contained 0,5–1 dose of macroelements and one dose of microelements, vitamins (10 mg/l of thiaminum, 1,0 mg/l of pyridoxine, 1,0 mg/l of nicotinic acid and 1,0 mg/l of ascorbic acid) supplemented with amino acids (250 mg/l of glutamic acid, 3 mg/l of tyrosine, 3 mg/l of arginine, 2 mg/l of hydroxyproline), plant growth regulators [0,5–1,0 mg/l of GA (gibberelline acid), 0,2 mg/l of 6-BAP (6-Benzylaminopurine, 0,1 mg/l of NAA (a-naphtylacetic acid)] in different variations. After seed germination, buds emerging and sprouts formation 1–2 cm in height, for propagation purpose they were passivated on the medium of other composition that differed from previous one by the content and ratio of growth regulators, especially by a high concentration of cytokinins [6-BAP (0,4–0,5 mg/l), kinetin (0,5 mg/l), adenine (0,5 mg/k)] in different variations in presence of GA (0,2 mg/l). In order to stimulate rhizomes growth, microclones were transferred on media with other composition and ratio growth regulators (6-BAP (0,2–0,3 mg/l) + GA (0,5–1,0 mg/l) or 6-BAP (0,2–0,3 mg/l) + GA (0,5–1,0 mg/l) + NAA (0,1 mg/l), in other words, with a high content of gibberellins. After the formation of rhizomes 10–15 cm in length, miscanthus plants were planted out in the open ground. Stimulation of rhizomes initiation and elongation on appropriate nutrient media before Miscanthus giganteus, M. sacchariflorus and M. sinensis planting in vivo resulted in 100% adaptation and 100% survival of plants in the winter period without the use of greenhouse complexes.
Conclusions. The method of miscanthus propagation in vitro and adaptation in the open ground was developed that included stimulation of rhizomes growth and favoured the increase of their length on media supplemented with gibberelline that guaranteed 100% preservation of microplants to be propagated from in vitro culture during adaptation in the open ground and acclimatization in winter. |
| Identification traits of secondary triticale genotypes for the use in breeding and plant growing | Author : Tetiana ?oskalets; Igor Hrynyk; VitaliyMoskalets; Valentyn Moskalets | Abstract | Full Text | Abstract :Purpose. To create and study genotypes of the secondary triticale of the hexaploid level for their effective use in further breeding and plant growing.
Methods. Field study, laboratory analyses, intraspecific hybridization with subsequent individual selection.
Results. New genotypes of the secondary triticale of the hexaploid level have been created and characterized for economic characters and agroecological traits and properties. Series of short-stem winter triticale was represented by ‘Pshenychne’, ‘Chaian’ to be adapted to the conditions of both intensive and organic farming. They are characterized by high drought resistance and winter hardiness, resistance to lodging, grain shedding, grain germination in the spike and spike fragility as well as by immunity to fungal diseases providing a high level of yield and technological quality of grain. The following new constant forms of triticale as ‘??_1-12’, ‘??_2-12’, ‘??_3-12’, that have an average height of the stem and belong to the Polissia-Forest-Steppe and Forest-Steppe ecotypes, demonstrated high productivity and adaptability in organic farming, particularly in case of the use of biologized elements of agrotechnology.
Conclusions. For the creation a new parent material during breeding of hexaploid triticale, the method of intraspecific hybridization is desirable with the use of parent material to be contrasting for eco-geographical origin and adapted local forms, followed by individual selection of genotypes with the desired characteristics and properties in cleavable hybrid populations. New genotypes of the secondary triticale have been created and characterized for breeding, genetic and agroecological traits and properties. In breeding practice, it is advisable to use a whole new approach of agroecological and genetic certification of genotypes for the effective solution of a number of theoretical and practical tasks facing modern ecological and adaptive breeding. New parent material of the secondary winter triticale and scientific support for its cultivation is proposed for further breeding and plant growing. |
| Rice (Oryza sativa L.) blast resistance genes bioinformatic analysis | Author : Katerina Bondarenko; Heorhii Slishchuk; Nataliia Volkova | Abstract | Full Text | Abstract :Purpose. To investigate rice blast resistance genes polymorphism by using bioinformatic methods.
Methods. Global and local nucleotide alignment, phylogenetic analysis, HyPhy test.
Results. For Pib gene, numerous single nucleotide substitutions and deletions of 1–3 bp were established. The phylogeny of this gene has been studied and homologues have been found both in various rice species and in other cereals. These sequences can encode proteins that «recognize» the phytopathogens effectors, and can also be associated with resistance to phytopathogens. The Pi4 gene is characterized by single nucleotide substitutions, insertions and deletions; the number of non-synonymous substitutions exceeds the number of synonymous ones. The Pi54 gene variability is significantly lower than that of the Pi4 and Pib genes. The predominant types of polymorphism were single nucleotide substitutions and small-sized indels. It was found that non-synonymous substitutions in Pi54, Pi4 and Pib genes were in close proximity, sometimes forming clusters, while some coding regions were either superconservative or contained predominantly synonymous substitutions. On philodendrograms, cultivated rice samples were clustered with samples of ancestral wild-growing species.
Conclusions. Evolution of the rice blast resistance genes Pi4, Pib and Pi54 is characterized by diversification selection. Considering that tense coevolution and significant rate of adaptation and creation of new pathogen races are typical for a plant and a parasite, these genes are subjected to intensive selection aimed at increasing diversity for obtaining the resistance to new races of the pathogen.

| Application of SSR markers to differentiate new varieties of soybean (Glycine max (L.) Merr.) | Author : Larysa Prysiazhniuk; Serhii Melnyk; Yuliia Shytikova; Iryna Sihalova; Alla Ivanitskaya | Abstract | Full Text | Abstract :Purpose. To study the molecular genetic polymorphism in new soybean varieties using SSR-markers as for the possibility to apply it for examination of varieties for difference, uniformity and stability.
Methods. Molecular genetic analysis of nucleic acids, cluster analysis.
Results. The results of study of molecular genetic polymorphism in 25 varieties of soybean through microsatellite markers using polymerase chain reaction were presented. Analysis of the amplification products has showed that for all the studied SSR-markers intra-species polymorphism was observed in the ‘Alinda’ variety, for the loci Satt 228 and Satt 726 – in the ‘Arnica’ variety, for the loci Satt 063 and Satt 726 – in the ‘Furio’ variety, which was taken into account in further studies of some genotypes. When assessing polymorphism in the studied varieties, it was determined that the frequencies of the identified alleles were ranging from 0,02 to 0,1 that depended on the microsatellite locus. Using cluster analysis, genetic distances were determined between varieties. The distance between many varieties, particularly in 59 cases, was 3.61, 3.16 and 2.83, these values of genetic distances prevailed in the studied sample of varieties.
Conclusions. Analysis of molecular genetic polymorphism showed that 10 varieties were the most polymorphic among 25 studied ones, that should be taken into account in their further identification. It was determined that the identified alleles were evenly represented in the sample of studied soybean varieties, as evidenced by a high polymorphic index of the locus (0.83–0.94). According to the evaluation of genetic distances between varieties, it was found that the varieties were the most similar by loci when the genetic distances between them were 2.00, and the varieties were the most different when the distances were 3.87. Thus, the marker system, which consists of four microsatellite loci such as Satt 063, Satt 114, Satt 228 and Satt 726, is effective for defining the difference between studied soybean varieties. |
| Screening of winter triticale genotypes for resistance to salinity in the shoot apical meristem culture | Author : Serhii Pykalo; Oksana Dubrovna | Abstract | Full Text | Abstract :Purpose. To conduct in vitro screening of different genotypes of winter triticale for resistance to salinity in the shoot apical meristem culture.
Methods. Plant tissue culture in vitro, in vitro breeding, statistical analysis.
Results. It was found that the increase of sodium chloride concentration from 0.6 to 1.5% resulted in inhibition of the callus culture growth in all genotypes that was indicative of the toxic effect of the stress factor. It turns out that 1.2% sodium chloride concentration allowed to differentiate triticale genotypes for salt tolerance. The line ‘38/1296’ appeared to be the most resistant to salinity stress because under breeding conditions calli of this genotype were characterized by higher morphogenetic potential, had the highest crude mass increase, and plants-regenerants were obtained only from explants of this line after cultivation on the medium containing 1.5% sodium chloride. The ‘ADM 11’ variety was the most sensitive to saline stress as mass necrosis and lack of regenerative ability in its calli were observed under breeding conditions. In the studied forms, genotypic dependence of morphogenesis processes in vitro culture was registered. From the induced calli, plants-regenerants were obtained, and their completion of growing, root development and transfer to in vivo conditions were optimized.
Conclusions. Genotypic response to salinity stress in the culture of shoot apical meristems of winter triticale was expressed by various crude mass increase and different morphogenetic potential on exposure to a stress factor. The line ‘38/1296’ can be used as a valuable material for further breeding of winter triticale. The culture of shoot apical meristems is recommended to apply as a test system for screening of triticale genotypes for resistance to salinity stress |

| Prospects of domestic viticulture transition to ecological (adaptive) management | Author : Irina Kovalyova; Lyudmila Gerus; Mariya Bankovskaya; Marina Fedorenko; ?lena Saliy | Abstract | Full Text | Abstract :Purpose. To determine the possibility of the domestic viticulture transition to the ecological (adaptive) management system, based on the use of highly adaptive varieties.
Methods. Procedures are conventional in viticulture. All stages of agrobiological research were carried out according to the M. A. Lazarevskiy technique. The resistance of genetic resources to fungal diseases was assessed using the 9-point scale on the natural infectious background with maximum damage. Organoleptic evaluation of wine and fresh grapes was performed using 8-point and a 10-point scales respectively.
Results. The main agrobiological traits (resistance to diseases, productivity and yield quality) of 22 perspective grape varieties and forms were studied. The level of resistance to fungal diseases was determined as one of the main criteria of the variety suitability for ecological (adaptive) viticulture. During five years, the average resistance level of this group of table and wine varieties was not lower than the relative one (6.5–7 points), and in ‘Zagrei’ variety – up to 7,5 points. The level of quality characteristics of products of new grape varieties and forms was determined (wine evaluation, marketability and assessment of fresh table grapes). Samples with a combination of high wine and grape quality characteristics and high productivity were selected. To replenish the gene pool of grapes by adaptive varieties, such perspective hybrid combinations as ‘Opalovyi’ ´ ‘Burmunk, ‘Avgustin’ ´ ‘Oryhinal and ‘Ohoniok tairovskyi’ ´ ‘Kardyshakh’ were studied. The level of group resistance to fungal diseases was identified and the perspective genotypes were preliminarily selected.
Conclusions. The level of display of a number of economic characters in the group of perspective table and wine grape varieties and forms was determined. Highly adaptive and highly productive varieties and forms suitable for the use in the adaptive viticulture were defined. Prospective hybrid combinations were considered, from which highly adaptive varieties will be selected as suitable for the ecological system of viticulture. |
| Seed quality of winter wheat varieties after black fallow depending on organo-mineral fertilizer application in the Left-Bank Forest-Steppe zone of Ukraine | Author : Sergii Avramenko | Abstract | Full Text | Abstract :Purpose. To determine the effect of the basic organo-mineral fertilizer on the formation and stability of grain quality of winter wheat varieties in multiple crop rotation after black fallow as a predecessor.
Methods. Field experiments were based on a multifactorial scheme using split-plot method with due regard to all requirements of the field experiment procedure, analysis of variance was used for statistical processing of the obtained results.
Results. Investigation data was given concerning determination of grain quality indices in winter wheat varieties of different ecotypes after black fallow as a predecessor depending on organo-mineral fertilizer application in the Left-Bank Forest-Steppe zone of Ukraine. In average for the period of investigation (2011–2015), the highest protein content in winter wheat grains was formed in no treatment variant [in such varieties as ‘Doridna’ (14.1%), ‘Dykanka’ (14.3%) and ‘Levada’ (14.2%)] and in case of organo-mineral fertilizer application [in the varieties ‘Hordovyta’ (14.0%), ‘Kalyta’ (14.0%), ‘Dykanka’ (14.7%) and ‘Levada’ (14.6%)]. The highest content of crude gluten in grains, without regard for the variant of the experiment, was found in the following varieties as ‘Dykanka’ (24.9–25.1%) and ‘Levada’ (23.7–25.4%).
Conclusions. It was established that the content of protein and crude gluten in grains as well as the falling number of winter wheat was highly dependent on such factors as the variety and the year of cultivation as compared to the fertilizer background. The following varieties as ‘Hordovyta’, ‘Mulan’, Dykanka’ and ‘Levada’ were very sensitive to the application of organo-mineral fertilizer for the protein content, while ‘Hordovyta’ (2.4%), ‘Levada’ (1.7%), ‘Borvii’ (1.2%) and ‘Mulan’ (1.1%) – for the crude gluten content. |
| Dependence of plant biometrics of cutting lettuce (Lactuca sativa L.) varietis on the concentration of microfertilizer Avatar-1 | Author : Vera Kutovenko; Nataliia Kostenko; Mykola Baranez | Abstract | Full Text | Abstract :Purpose. To investigate the variability of plant biometrics of cutting lettuce (Lactuca sativa L.) varieties depending on the concentration of microfertilizer Avatar-1.
Methods. Field study, biometric technique, comparative approach, statistical evaluation, generalization.
Results. Dependence of the plant height, the diameter of the leaf rosette, the number of leaves per plant, the leaf area of plants on the concentration of microfertilizer Avatar-1 was defined. Investigations of cutting lettuce ‘Afitsyon’ and ‘Concord’ varieties by Dutch breeding were conducted in 2016–2017 in the collection sites of the department of vegetable growing in the scientific-experimental field “Fruit and vegetable garden” of the National University of Life and Environmental Sciences of Ukraine. In order to determine dependence of plant biometrics of cutting lettuce varieties on the concentration of complex microfertilizer Avatar-1, the following scheme was used for the both varieties: variant 1 – water (control); variant 2 – 0,10% solution; variant 3 – 0,25% solution; variant 4 – 0,50% solution. Plants were treated with microfertilizer three times during the vegetative period. Alterations of plant biometrics of cutting lettuce ‘Afitsyon’ and ‘Concord’ varieties depending on the concentration of complex microfertilizer Avatar-1 was studied.
Conclusions. It was found that in case of three-time plant treatment with complex microfertilizer Avatar-1 at the time of harvesting, the best plant biometrics was registered in variant 3 (concentration 0,25%) for the both ‘Afitsyon’ and ‘Concord’ varieties of cutting lettuce. The height of plants of the cutting lettuce in ‘Afitsyon’ variety exceeded this figure in ‘Concord variety by 1,1–1,4 cm. The concentration of microfertilizer had no significant effect on the diameter of the leaf rosette of ‘Concord’ variety (25,1–25,9 ??). The diameter of the leaf rosette of ‘Afitsyon’ variety was 26,0–28,7 cm. In cutting lettuce, the largest leaf area per plant was registered in ‘Afitsyon’ variety (3516.5 cm2/plant ) in case of plants treatment with 0,25% solution of complex microfertilizer Avatar-1 that exceeded this figure in ‘Concord’ variety by 660 cm2/plant. The best plant biometrics of cutting lettuce of the studied varieties and optimal concentration of complex microfertilizer Avatar-1 (0.25% solution) was defined that is important for improving agricultural cultivation techniques. |
| Practical aspects of applying statistical analysis of quantitative characters of ?utting lettuce varieties var. ?apitata L. | Author : Nadiya Leschuk; Nataliya Orlenko | Abstract | Full Text | Abstract :Purpose. According to the duration of interphase periods, it can be noted that such varieties as ‘Dumka’ and ‘Dyvohrai’ had the highest rate of maturation in comparison with ‘Bona’ and ‘Hodar’ varieties, and the lowest one as compared to the ‘Olzhych’ variety |
| Variability of seed quality indices in soft spring wheat depending on weather conditions | Author : Iryna Pravdziva; Nadiia Vasylenko; Svitlana Khomenko | Abstract | Full Text | Abstract :Purpose. To study the effect of weather conditions on grain quality in soft spring wheat lines that are undergoing competitive variety trial. To identify indices to be least affected by weather conditions. To investigate the correlation dependence between grain quality indices in the years with contrasting weather conditions and conduct the analysis of variance.
Methods. Parameters of grain and flour quality of bread spring wheat lines were determined using conventional methods at the laboratory of grain quality of V. M. Remeslo Myronivka Institute of Wheat, NAAS of Ukraine, protein content in flour was measured with the use of the SPECTRAN 119M device.
Results. Contrasting weather conditions were considered including arid ones (2013) – that had negative effect on 1000 kernel weight, but at the same time positively influenced the averaged grain quality indices; with excess humidity (2014) – they resulted in decreasing of all technological indices; optimal ones (2015) – allowed to obtain maximum indices of grain and flour quality. Paired coefficients of correlation between different parameters of grain and flour quality were calculated. The most number of strong and medium correlations was found in a dry year (33.3%), the smallest one – in a wet year (13.0%). Such indices as flour strength, dough dilution and protein content depended on the conditions of the growing year least of all. Grain-unit response was the most considerable to the humid conditions of the year. The analysis of variance showed that climatic conditions had significant effect on the quality indices of grain and flour. At the same time, an important genotypic component was revealed in indices of dough resilience (63%), bread volume and rating (61 and 53% respectively), the flour strength (42%), crude gluten quality (33%), bread porosity (30%). With climate variations, the genotypic conditionality of the content of protein and especially crude gluten was expressed insignificantly.
Conclusions. When creating high quality cultivars, plant breeders should be guided by such indices as flour strength, dough resilience, gluten quality, bread volume and rating as only they are more genetically determined |
